The isolation of a 20kb transposon, tn4527, carrying resistance to trimethoprim, spectinomycin and streptomycin

ABSTRACT

Tn4527 was isolated from a Salmonella typhimurium strain obtained in Brazil. Its size is approximately 20 kb, being one of the largest antibiotic resistance transposons isolated from Enterobacteria. It carries resistance to trimethoprim , spectinomycin and streptomycin. It transposed originally to R64drd11 and from this plasmid to pDS4161, a multicopy Cmr ColK derivative.
Cms derivatives were obtained from pDS4161: : Tn4527. The Cms isolates have lost a 1.6kb Pst I fragment, including DNA from the pDS4161.
The Tp resistance carried by Tn4527 is not expressed in one of the strains used (FG 151). 1 Tp: trimethoprim; Sp: spectinomycin; Sm: streptomycin; Cm: chloramphenicol; Km: kanamycin; Ap: ampicillin; Tc: tetracycline.
