Sister chromatid exchange in mutagenicity testing:a study of mercapto oxazolyl carboxylic acid

ABSTRACT

Sister chromatid exchange (SCE) and cell cycle kinetics were studied as methods for determining the toxic effects of chemicals, especially mutagens and carcinogens. A clastogenic substance, mercapto oxazolyl carboxylic acid, an anti-rheumatic drug, was added to human lymphocyte cultures treated by the BrdU-Giemsa technique. The drug induced increased SCE frequency at concentrations lower than those that induce chromosome aberrations. At the lowest concentrations, the drug caused earlier onset of the cell cycle, whereas at the highest concentrations used, the cell cycle was delayed.
On the basis of the present results, we conclude that SCE score and cell kinetics represent important and sensitive methods for analyzing drug mutagenicity and toxicity.
